Galactinol Synthase is an Extravacuolar Enzyme in Tubers of Japanese Artichoke (Stachys sieboldii).

چکیده
Galactinol synthase (GS, UDP-alpha-d-galactose:1l-myo-inositol-1-O- alpha-d-galactopyranosyltransferase) is a key enzyme in the biosynthetic pathway of the raffinose family of oligosaccharides. The subcellular location of GS was studied in the parenchyma of stachyose-storing tubers of Japanese artichoke (Stachys sieboldii) by isolation of protoplasts and vacuoles. A comparison of the activities of GS, malate dehydrogenase, and alcohol dehydrogenase (extravacuolar markers) and alpha-mannosidase and beta-N-acetylglucosaminidase (vacuolar markers) in parenchyma protoplasts with those of vacuoles isolated from them showed that GS was an extravacuolar enzyme.

Transport of Stachyose and Sucrose by Vacuoles of Japanese Artichoke (Stachys sieboldii) Tubers.
Vacuoles are the stores for large amounts of stachyose [alphagal (1,6) alphagal (1,6) alphaglc (1,2) betafru] in tubers of Japanese artichoke (Stachys sieboldii). The uptake of stachyose by these vacuoles was examined and compared with that of sucrose. The uptake mechanisms of both sugars were quite similar. The kinetics showed a single saturable response to increasing external concentrations o...
متن کاملGalactinol as marker for seed longevity.
Reduced seed longevity or storability is a major problem in seed storage and contributes to increased costs in crop production. Here we investigated whether seed galactinol contents could be predictive for seed storability behavior in Arabidopsis, cabbage and tomato. The analyses revealed a positive correlation between galactinol content and seed longevity in the three species tested, which ind...

The effect of photoperiod and growing degree days (GDD) on dry matter and dry matter partitioning in Jerusalem artichoke was investigated during 2008-09 and 2009-10. Three Jerusalem artichoke genotypes (CN-52867, JA-89 and HEL-65) were planted in 15 day-intervals between with thirteen different dates (September 20 to March 20) atKhon Kaen University,Thailand. Jerusalem artichoke genotypes ...
متن کاملToxicity of Carbon Tetrachloride in Japanese Quails: Evaluation the Effect of Artichoke (Cynara scolymus) Powder on Performance and Immune Response
This study was conducted to investigate the effects of artichoke powder on performance and immune function of Japanese quails poisoned with carbon tetrachloride (CCl4). One hundred meat type (1-d-old) quails were assigned randomly to 4 treatment groups (n=25). Treatment groups included with the study were: 1) control group (without artichoke supplementation or CCl4 administration, 2) group with...
متن کاملAnalysis of the raffinose family oligosaccharide pathway in pea seeds with contrasting carbohydrate composition.
Raffinose family oligosaccharides (RFOs) are synthesized by a set of galactosyltransferases, which sequentially add galactose units from galactinol to sucrose. The accumulation of RFOs was studied in maturing seeds of two pea (Pisum sativum) lines with contrasting RFO composition. Seeds of the line SD1 accumulated stachyose as the predominant RFO, whereas verbascose, the next higher homolog of ...
